دراسة تحسين انحلاليّة السلفاسالازين بتأثير بعض محسّنات الانحلاليّة

Abstract

تمّت دراسة تأثير مادة (β-) سيكلودكسترين على انحلاليّة السلفاسالازين-الدواء المستخدم في علاج التهاب الكولون القرحي-في الماء في ثلاث درجات حرارة مختلفة (25˚، 35˚، 45˚)، وأظهرت منحنيات الانحلاليّة زيادة في انحلاليّة المادّة في الماء بزيادة تركيز الـ (β- سيكلودكسترين)، وبارتفاع درجة الحرارة. وقد تمّ تحضير معقّد الاندماج بين السلفاسالازين والـ (β- سيكلودكسترين) بطريقة البلورة، وحُضِّرت مضغوطات تتضمن هذا المعقد. كما أُجريت مقارنة لمعدّل ذوبانها مع المضغوطات الحاوية على المادة لوحدها وتمّت ملاحظة سرعة الانحلال المرتفعة مقارنةً مع مضغوطات المادة. وأُجريت دراسة لمراقبة تأثير المواد الأخرى المساعدة على الانحلال السلفاسالازين كالتوين 60، وهيدروكسيبروبيل (β- سيكلودكسترين) بالمقارنة مع الـ (β- سيكلودكسترين) في درجة حرارة 25˚؛ فتبيّن أنّ الزيادة التي يُحدِثها (التوين 60) في الانحلاليّة أقل ممايحدثه الـ (β- سيكلودكسترين) باستخدام نفس التركيز. أَمّا هيدروكسي بروبيل (β- سيكلودكسترين) فقد أحدث زيادة ملحوظة في الانحلاليّة مقارنةً مع الزيادة التي يسببها
الـ (β- سيكلودكسترين) في درجة الحرارة ذاتها.

Sulfasalazine, a drug used to treat ulcerative colitis is a poorly water-soluble. The effect of β-cyclodextrin on the aqueous solubility of sulfasalazine was investigated in three different temperatures 25˚, 35˚, and 45˚. Phase-solubility profile indicated that the solubility of sulfasalazine was significantly increased in the presence of β-cyclodextrin by increasing cyclodextrin concentration and temperature. Solid complexes were prepared by crystallization method.  Vitro studies showed that the solubility and dissolution rate of the drug were significantly improved by complexation with β-cyclodextrin with respect to the drug alone. The effect of other cosolvents like tween 60, and hydroxyl propyl  β-cyclodextrin (HP β-CD) was also investigated in 25˚.The  solubility induced by tween 60 was less than that induced using β-cyclodextrin, whereas HP β-CD caused better sulfasalazine solubility  with respect to β-cyclodextrin in the same temperature.
