تحرّي تعبيريّة البروتين P53 في الغدّة النكفيّة المعرّضة لإشعاع الأمواج الميكرويّة الصادرة عن الهاتف الجوّال
Abstract
تم إجراء البحث بهدف دراسة إمكانيّة حدوث طفرات في المورّثة Tp53 في الغدّة النكفيّة بعد تعريضها لإشعاع الأمواج الميكرويّة، عن طريق دراسة مستويات البروتين P53.
تألّفت عيّنة البحث من (16) أرنباً, مُقسَّمة بالتساوي الى مجموعتين: شاهدة وتجريبيّة وُفقاً للتعرّض لأشعّة الأمواج الميكرويّة، حيث عُرِّضَت المجموعة التجريبيّة لإشعاع الهاتف الجوّال مدة ساعة واحدة يومياً، (6) أيام في الأسبوع، لمدة (6) أشهر. وبعد انقضاء فترة التعريض تمّت التضحية بالأرانب واستئصال الغدّة النكفيّة، وبالتالي تم الحصول على (16) خزعة نسيجيّة مُلوَّنة مناعياً بالضد وحيد النسيلة P53، وتم استخدام طريقة عد الخلايا الإيجابيّة في كل ساحة نسيجيّة، وثم حساب النسبة المئويّة لها من مجموع خلايا الساحة المدروسة عند التكبير العالي ×400.
أبدت جميع العيّنات تعبيريّة سلبيّة تجاه الملوّن P53, أي عدم حدوث طفرات في المورّثة TP53 بسبب عدم التعبير عن البروتين الطافر P53.
The purpose of the research is to study the possibility of mutations in the Tp53 gene in the parotid gland after exposure to microwave-radiation, by studying the levels of P53 protein.
The sample consisted of 16 rabbits divided equally according to exposure to microwave – radiation into two groups: control and experimental, where the experimental group was exposed to the mobile phone one hour daily – 6 days a week – for 6 months.
After the end of the exposure period, rabbits were sacrificed and parotidectomy was removed, and thus we obtained 16 tissue biopsies immunohistochemically stained with the monoclonal antibody P53, and the method of counting positive cells was used in each field of tissue and then calculated the percentage of them from the total cells of the field studied at high magnification 400×.
All samples were negative for stained P53, i.e. no mutations occur in TP53 gene due to non-expression of mutant P53 protein.
