النشاط الصاد للجراثيم والخصائص المورفولوجية والفيزيولوجية والحيوية الكيميائية لعزلة الجنسStreptomyces من المياه البحرية لمدينة اللاذقية
Abstract
تمّ الحصول على جراثيم Streptomyces من المياه البحرية لمدينة اللاذقية في موقع أفاميا فقط وزرعت العينات على وسط Marine-Agar medium ووسط Starch Casein Agar ووسط Actinomyces Isolation Agar، وتميزت مستعمراتها بأنها ذات أقطورة هوائية بلون أبيض وأفطورة ركائزية منغرسة ضمن الوسط بلون زهري وسلاسل الأبواغ حلزونية لا تنتج أصبغة منحلة ولا أصبغة الميلانين على وسط peptone-yeast extract-iron agar. أشارت نتائج دراسة الخصائص المورفولوجية والفيزيولوجية والزرعية والحيوية الكيميائية لعزلة Streptomyces وفقا لدليل بيرجي أنها تعود إلى النوع Streptomyces rochei . أظهر النوع المعزولStreptomyces rochei خاصية التصادية الجرثومية تجاه جميع الجراثيم الممرضة البشرية السالبة صبغة غرام (Escherichia coli, Klebsiella pneumoniae, Serratia marcescens, Pseudomonas aeroginosa, Proteus vulgaris)، والموجبة صبغة غرام (Streptococcus faecalis, Staphylococcus aureus, Staphylococcus albus) بتقانة الزرع المتعامدة، وتراوحت أقطار حلقات التثبيط لمستخلص النوع Streptomyces rochei بالمذيب خلات الإيتيل بتقانة الانتشار بالأقرص ضمن المجال 10 ملم و 34.6 ملم تجاه جراثيم Escherichia coli و Staphylococcus albus على التوالي. Bacteria of Streptomyces was isolated from sea water of Latakia at Afamia station only, by using Marine-Agar medium, Starch Casein Agar medium, Actinomyces Isolation Agar, their colonies were white aerial mycelia and pink substrate mycelia, spores were spirals, didn't produce soluble pigments and melanin on Peptone-Yeast Extract-Iron Agar. Results of morphological, physiological cultural, biochemical characteristics indicated that the Streptomyces isolated according to Bergey's manual of determinative bacteriology belongs to the species Streptomyces rochei. Streptomyces rochei showed antibacterial activity against all human pathogenic Gram negative bacteria (Escherichia coli, Klebsiella pneumoniae, Serratia marcescens, Pseudomonas aeruginosa) and Gram positive bacteria (Streptococcus faecalis, Staphylococcus aureus, Staphylococcus albus) by cross streak method. Inhibition cycles of ethyl acetate extract of Streptomyces rochei by disc diffusion method ranged between 10 mm and 34.6 mm against Escherichia coli and Staphylococcus albus respectively.Downloads
