دراسة تأثير بعض منظمات النمو في الإكثار الدقيق لنبات العرن المثقب Hypericum perforatum

Abstract

detailed in vitro multiplication system for rapid micropropagataion of the Hypericum perforatum L, has been developed by using  tissue culture technique . The  shoots (1- 0.5cm) were cultured on  MS ( Murashige and Skoog 1962) then placed onto  based medium containing a combination of growth regulators at different  concentrations (BA , Kin , IBA ) . After that they were placed into MS based medium containing NAA and IBA.   Results showed that the highest number of growth was 39.6 when ( 0.6 mg/l BA + 0.1 mg/l IBA)  were adding (MS3), also the highest of the growth was (3,81cm) in medium MS3 .The observed rooting was (7.1roots , 10,5 cm ) length and %95 in percentage of rooting) in R5 (MS+1mg/l NAA) , and acclimatization was 85%

 

تم تطوير تقنية لإكثار نبات  Hypericum  perforatum في المختبر بإستعمال تقنية  زراعة النسج النباتية من خلال عقل بطول (1- 0.5) سم والتي زرعت على وسط (Murashige and Skoog ,1962)    ( MS)وبعد ذلك تم إكثارها على وسط MS المضاف إليه منظمات نمو مختلفة ( البنزيل أدنينBA ،الكينتين Kin وإندول بيوتريك أسيد IBA ( حمض الزبدة )  ) . أوضحت النتائج أن أكبر نسبة لمتوسط  النموات المتشكلة كانت 39.6 في المعاملة MS3 التي تحتوي IBA ( 0.1 mg/l )  + BA     ( 0.6 mg/l )  وكان  أكبر  طول للنموات أيضاٌ على المعاملة   MS3حيث بلغ أكبر طول للنموات 3.81  سم . تمت بعد ذلك عملية التجذير على أوساط  MS  المضاف إليها IBA أو NAA ) نافتيل حمض الخل (و كانت المعاملة R5 التي تحتوي على NAA  (mg/L  1) هي الفضلى حيث بلغت  نسبة التجذير %95  و متوسط عدد الجذور  7.1 سم , ومتوسط طولها 10.5 سم . بلغت نسبة الأقلمة %85 من عدد النموات التي أمكن الحصول عليها.
