التباين الوراثي في القمح الطري باستخدام بروتينات التخزين
Abstract
يهدف هذا البحث إلى دراسة التباينات الوراثية الموجودة في بعض الطرز الوراثية المختلفة من القمح الطري باستخدام الرحلان الكهربائي البروتيني للغليادين والغلوتينين. وقد استخدم في هذه الدراسة (10) طرز وراثية من القمح الطري, وكان عدد الحزم البروتينية الناتجة عن تحليل الغليادين وفق تقانةA-Page (Electrophoresis Acidic Polyacrylamide Gel) 13)) حزمة متباينة. بينما أظهرت نتائج تحليل الغلوتينين ذات الوزن الجزيئي المرتفع وفق تقانة Sodium Dodecyl Sulphate Polyacrylamide Gel Electrophoresis((SDS-Page إلى وجود الحزمتين null و2* في الموقع Glu-A1 و الحزم 17+18,7,7+8 في الموقع Glu-B1 , فضلاً عن الحزم 5+10,2+12 في الموقع Glu-D1 و تم تحديد الطرز االوراثية التي تمتلك مواصفات تكنولوجية عالية.جُمعت البيانات الناتجة عن كلا التقانتين، ورسمت شجرة القرابة الوراثية التي توضح التشابه الوراثي بين الطرز الوراثية المدروسة وفق معادلةDice بطريقة المتوسط الحسابي للمجموعات الزوجية غير المزانة Unweighted Pair Group Mean Arithmetic Average (UPGMA).كما أظهرت النتائج أن متوسط نسبة التشابه الوراثي بين الطرز الوراثية المدروسة قد بلغت 71% و تميزت بعض الطرز الوراثية الأخرى بأعلى نسبة تشابه وراثي بينهما, إذ بلغت %100.
The genetic variation within different genotypes of wheat was studied by proteins electrophoresis glutenin and gliadin. In this study 10 genotypes bread wheat were characterized. A Number of protein bands were detected 13 polymorphic bands for Electrophoresis) gliadins analyses .The results of A-Page( Acidic Polyacrylamide Gel SDS-PAGE (Sodium Dodecyl Sulphate Polyacrylamide Gel Electrophoresis) indicate that subunits /alleles 2* and null at Glu-A1, 7, 7+8 and 17+18 at Glu-B1, 2+12 and 5+10 at Glu-D1, and identify the genotypes that have good technology characteristics. Data were combined together to calculate the genetic similarities between studied individuals using Dice coefficient followed by setting up the cluster analysis using Unweighted Pair Group Mean Arithmetic Average (UPGMA) method. The value of average genetic similarity was 71% between varieties, and The value of genetic similarity in some genotypes was 100%.
