استخدام الاستخلاص بالأطوار الصلبة لتحسين الفصل الكروماتوغرافي للمكونات الفعالة في الأشكال الصيدلانية المتباينة التركيب
Abstract
بدأت تقنية الاستخلاص بالأطوار الصلبة في تحضير العينة تحل محل الاستخلاص التقليدي سائل – سائل لما لها من ميزات إضافية.
تضمَّن البحث تعريفاً بمبدأ وطريقة استخدام هذه التقنية إضافة إلى استخدامها في حل مشكلة المراقبة الكيميائية لمكونات أحد الأشكال الصيدلانية متباينة التركيب حيث تحتوي المضغوطة الواحدة على /500/ملغ من الباراسيامول وعلى كميات زهيدة من فوسفات الكودئين وماليات الكلورفينيرامين وكلورهيدرات الافيدرين.
مكّن الاحتجاز القوي للمركبات الزهيدة على الطور الصلب – خلافاً للمكون الرئيسي (الباراسيتامول) من عزله عن باقي المكونات إضافة إلى زيادة تركيز هذه المكونات في محل الاستخلاص الأخير.
تم فصل المكونات المستخلصة كروماتوغرافياً باستقلالية جيدة وباستخدام طور متحرك واحد وتم كشف المركبات المفصولة بمتحري الأشعة فوق البنفسجية.
استخدم الفصل الكروماتوغرافي أساساً لطريقة تحليلية سهلة التطبيق صحيحة تفيد في المراقبة الكيميائية لمكونات الشكل الصيدلاني للحكم على نجاح عملية التصنيع في تأمين تجانس صحيح للمكونات الزهيدة في أرجاء الشكل الصيدلاني الصلب.
Solid phase extraction (SPE) is a new technique that substitutes the traditional liquid-liquid extraction for sample preparation.
In Addition to definition of the principle, utilization and advantage of (SPE)a new analytical application has been dealt with.
SPE is used to resolve the chemical control of multi ingredients heterogeneous dosage form.
One tablet contains 500 mg of paracetamol, 5mg of chlorpheniramine maleate, 8mg of codeine phosphate and 2mg of Ephedrine hydrochloride.
The strong retention of the minor ingredients on SPE is used to isolate and concentrate them in the final extracting solvent.
The extracted components are separated with good resolution by isocratic elution chromatography and detected by U.V. detector.
The proposed chromatographic method is convenient for judging the homogenous dispersion of ingredients in the dosage from.
