دراسة إنتاش حبوب الطلع ونمو الأنابيب الطلعية في الزجاج عند نبات الحوذان Ranunculus Circinatus Sibth.
Abstract
درسنا مجموعة من العوامل الهامة التي تلعب دوراً كبيراً في عملية انتاش ونمو الأنبوب الطلعي في نبات Ranunculus circinatus وهذه العوامل هي السكروز بتراكيز عدة (50-75-100-125غ/لتر) وتأثير الحرارة بدرجات عدة (20-25-30°م)، ودرجة الحموضة بدرجات عدة (4، 5، 6، 7)، وتأثير البور والكالسيوم وكذلك الفترة الزمنية المثالية للحصول على أفضل نمو للأنبوب الطلعي وأعلى نسبة للانتاش.
أظهرت الدراسة أن وجود الكالسيوم والبور في الوسط الأساسي ضروري لحادثة الانتاش ونمو الأنبوب الطلعي. إذ لم يحدث انتاش في غياب الكالسيوم وكانت نسبة الانتاش قليلة في غياب البور (4%) ولم يتجاوز طول الأنبوب الطلعي (150 ميكرون).
أما أعلى نسبة انتاش تم الحصول عليها (88%) كانت بعد 3 ساعات من الزراعة وأفضل نمو للأنبوب الطلعي على الوسط الأساسي (715 ميكرون) عند درجة الحرارة 25°م وعند PH = 6 وبوجود 100غ/ليتر من السكروز.
A group of important factors that played a great role in germination and growth process of androecium tube in R. circumatus were studied, these factors are: sucrose at many concentration (50, 75, 100 and 125 g/l), influence of temperature at various degrees (4, 5, 6 and 7&), and PH at different degrees (4, 5, 6 and 7), in addition to, the impact of Bore (B) and calcium (Ca). moreover, it is the optimum temporal period to obtain ratio of germination.
This study revialed that the presence of B and Ca in the basal medium was necessary for process of germination and growth of androecium tube, therefore, the germination did not take place in absence of Ca and the germination ratio was low in absence of B (4%). the ratio did not overstep the long of androecium tube (150 um).
While, the highest ratio of germination was obtained (88%) after three hours of culture, and the best growth of andoecium tube was in the basal (or original) medium (715um) at 25°C and PH = 6 and presence of 100g/l sucrose.
