استخدام تقانة الدنا (DNA) المضخم عشوائيا RAPD لكشف درجة القرابة بين بعض أنواع جنس الحوذان Ranunculus, L في الساحل السوري
Abstract
تهدف هذه الدراسة الحصول على معلومات حول القاعدة الوراثية ودرجة القرابة بين طرز وراثية من جنس الحوذان Ranunculus, L. موزعة على الشريط الساحلي في سوريا باستخدام تقانة الدنا (DNA) المضخم عشوائيا RAPD. استخدم 27 مرئسة (primer) استطاع 16 منها إظهار درجة مختلفة من التعدديات الشكلية التي قسمت الطرز المدروسة استنادا إلى نتائج التحاليل بالمتوسط الحسابي للمجموعات الزوجية غير المزانة UPGMA والنسبة المئوية لدرجة عدم التوافق Percent disagreement إلى عنقودين رئيسين ضم العنقود الأول الأنماط الوراثية الرطبة و ذات لون التويج الأصفر و شكل الأوراق المجزأ، بينما تضمن العنقود الثاني الأنماط الوراثية المائية وذات التويج الأبيض و شكل الأوراق الخيطي. توافقت هذه النتائج المستحصل عليها بتقانة RAPD بشكل كامل مع جميع البيانات المتوفرة المستندة إلى دراسة الصفات المورفولوجية والتوزع البيئي لهذه الطرز.
This study seeks to obtain information about genetic base and relationship degree between genotypes belonging to genus Ranunculus distributed at coastal line in Syria by using RAPD technique. Twenty seven primers were used, sixteen primers exhibited deferent degree of polymorphism’s which divided studied types according to results of analysis by UPGMA and percentage of disagreement degree into two main clusters: The first cluster includes hygrogenotypes (or: wet genotypes) and with yellow corolla and divided (or parted) leaves form (or :morph), while, the second cluster includes genotypes with white corolla and fellamental (or: thread) leaves form.
These results which are obtained by RAPD technique completely agreed with all available data depending on studying morphological characters and environmental distribution for these types.
