تأثيـــــر تركيــــب الوســـط المغــــذي فـــي إنبــــات ونمـــو بـــذور نبــــات الكحلاء (Echium angustifolium Mill.) المزروعـة مخبريــاً فـي الأنابيب
Abstract
لقــــــــــد تـــــــم فـــــي هـــــــــــذا البحــــــث دراســــــــــة تأثيـــــــــــر تركيـــــــــب الوســــــــط المغـــــــــذي فـــــــي إنبــات ونمــو بـذور النــوع Echium angustifolium Mill. (نبـــــات الكحــلاء) المزروعـــــــــــة مخبريــــــــــاً فــــــــي الأنابيب In vitro. تـــــم استخـــــــــدام الأوســـــــاط التاليــــــــة (المــــــاء، وســــــــــط MS جـــــامد Murashige & Skoog، وســط 1/2 MS جامد) بــــوجــــــود هرمــــــــون الجبريليـــــــن GA3 1-0.1-0.01-0.001) مغ/ل) أو بـــــــدونـــــــــه. أظهـــــرت النتــائـــــــج تفـــــــــوق الوســــــــط MS الجامـــــــــد بوجـــــود الجبريليــــــن 1مـــــــغ/ل علـــــــى باقـــــــــي التراكيــــــــز والأوســــــاط الأخــــــرى، حيث سجــــل أعلى نسبــــــة إنبـــــــات بمعـــــــدل 68.88%، وبمتوســـط طـــول جذيـــر بلـــــغ 26.3مـــــم، ومتوســـط طــــــول سويقــــــة 37مــــــم بعد أسبوعين مــــــــن الزراعـــــــــــــــة. كما تحسنت نسبة الإنبات في الوســــط 1/2 MSالجامـــــــد مضافاً إليه الجبريلين بتركيــــــــــز 1مــــــــــغ/ل، حيـــــــث بلغــــــت 64.43%، وســــــجـــل متوســـــــــط طـــــول الجذيــــــــر 24.5مـــــــم، والسويقــــــة 33مــــــــم. كما أدى إضافـــــــــــة الجبريليــــــــــن 0.001 مـــــــغ/ل إلـــــــــى المـــاء، إلى الحصول على إنبات 64% بعــــــد أسبوعيـــن مـــــــن الــــــــزراعة، وبمتوســــــــط طــــــــول جذيـــــــر 18 مـــــــم، ومتوســــــط طـــــــول سويقـــــــة 20 مــــــــم. زرعـــــــــــت بعــــض البـــــــادرات الناتجـــــــة عـــــــن أفضــــــل وســـــــط فــــــي أصص تحـــــــوي تربـــــــة مغذيــــــة (تورب) وأقلمتها، وتـــــــم الحصــــــول علـــــــى نباتـــــات بحالــــــة خضريــــــــة جيـــــــدة أخذت نتائجها بعد شهريــــــن مــــــن الزمـــــــن. The effect of different culture medias (water, MS solid, MS 1/2 solid) on germination and growth of Echium angustifolium Mill. in vitro was studied with different concentrations of gibberellic acid (0.001-0.01-0.1 and 1 mg/l). The results showed that best germination rate (68.88%) and root (26.3mm) and shoot (37mm) growth were obtained in MS solid medium with 1mg/l GA3. Addition of 1mg/l GA3 to MS1/2 solid medium improved germination rate (64.43%), root (24.5mm), and shoot (33mm)growth. When GA3 (0.001 mg/l) was added to water medium, germination rate reached (64%), as well as root and shoot growth (18mm and20mm) alternatively after 2 weeks of planting. Seedling of MS solid medium were transferred into pots contained torp medium to adapting them with outside environment, and then surviving their growth until maturity after 8 weeks.Downloads
